MOTS-C is a peptide whose instructions sit in mitochondrial DNA. Mitochondria are the structures inside cells that turn food and oxygen into usable energy, and they carry a small set of genes of their own.
Mitochondria have their own small set of genes, left over from when they were separate organisms billions of years ago. MOTS-C was discovered in 2015 hiding inside one of those genes.
That made it one of the first known examples of mitochondria writing their own protein messages to the rest of the cell, rather than only taking instructions from the nucleus.
Research since then has focused on metabolism and exercise. Levels appear to rise with physical activity, which is part of why it gets called an exercise-mimicking peptide.
Cells have an energy sensor called AMPK. When energy runs low, as it does during exercise or fasting, AMPK switches on and shifts the cell toward burning fuel rather than storing it.
In animal studies, MOTS-C activates that same sensor. That is the mechanism behind claims about metabolism, insulin response, and endurance.
Under stress it also travels to the cell nucleus and changes which genes get read. That is why researchers describe it as a message running backward, from mitochondria out to the rest of the cell.
The animal results are genuinely interesting. Human trials to confirm any of it are only starting.
